Restriction digests of the clone give the following sizes (kb): PstI--4.4, 0.54 + others; EcoRI--5.3; HindIII--5.3; BamHI--5.3; SalI--5.3. The insert contains HinfI, HpaII, PstI, HincII, SstI, and PstI sites. The PstI sites at the ends were created by the cloning procedure. This is a 5' end probe for CEA. Open reading frame encoded in pCEA1 and pCEA2 contains 2 closely-related repeats of 178 amino acids each. pCEA1 contains repeat a and part of repeat b; pCEA2 contains repeat b and part of repeat a. The 0.534 kb PstI fragment detects mRNA of 3.8 and 3.2 kb from mouse mammary tumors. |
