Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.65, 1.95; HindIII--2.65, 1.95; PstI--2.65, 1.75; SalI--2.65, 1.95; EcoRI--4.5. This probe was originally assigned to D22S11. However, it has since been shown that D22S11 is within the BCR locus. Insert is 1.95 kb BglII/HindIII fragment containing exon 1 given in fig. 2b. The BglII site was lost by subcloning. The insert contains 2 TaqI sites; the polymorphic one is 0.5 kb from the HindIII end. Depositor verified that this preparation detects the TaqI RFLP. Enzyme(s) not detecting polymorphism: RsaI, PstI, EcoRI. Tested in 12 unrelated middle-Europeans. |
