Restriction digests of the clone give the following sizes (kb): EcoRI--2.75, 2.6; BamHI--5.45; PstI--4.5, 0.52, 0.42; HindIII3.9, 1.55; XbaI--5.45. Contains a point mutation at position 183 (T to C) resulting in a Cys to Arg substitution. The mutant protein does not bind lymphocyte function antigen (LFA-1). Clones K4-1.1 (ATCC 63043) and K3-1.1 (ATCC 63044) differ in the 5' end of the insert. Nt 638 of the K3-1.1 insert equals nt 132 of the K4-1.1 insert. The insert has an open reading frame of 1611 nt and 29 nt of 5' untranslated sequence. It codes for a functional transmembrane protein. The overall protein structure is quite similar to human ICAM1 and there is 50% homology at the amino acid level. |
