The insert includes the following restriction sites (nucleotide positions according to GenBank K02782): PstI--2676, 3511, 3607, 4846; PvuI--4017; StuI--4041; SalI--4677; HindIII--5060. Restriction digests of the clone give the following sizes (kb): EcoRI--6.7; SalI--4.9, 1.8; PstI--4.3, 1.3, 0.8, 0.3; HindIII--5.9, 0.8. pMLC3/4 contains the 3' portion of the cDNA, encoding most of the alpha chain portion which begins at nucleotide 2067. The 5' end of the insert is between the StuI (2223) and PstI (2676) sites. |
