Insert contains the following sites separated by (bp)(approximate): EcoRI- 440- PstI- 750- KpnI- 910- EcoRI. Restriction digests of the clone give the following sizes (kb): EcoRI--3.5, 2.2; PstI--4.6, 0.52; KpnI--4.3, 1.23; HindIII--5.7; BamHI--5.7. There is 99.6% nucleotide homology between Prn-p of NZW mice (short scrapie incubation period) and I/LnJ mice (long scrapie incubation period). BstEII detects an AC to GT change at codon 189 which results in Val (I/Ln) or Thr (NZW). It also correlates with a T to C change at codon 108 which results in Phe (I/Ln) or Leu (NZW). XbaI, PstI and EcoRI RFLPs lie 3' to exon 2. All 3 inbred strains known to have long scrapie incubation periods lack the BstEII site at codon 189. Seventeen short and intermediate incubation period mice have this site. Isolated from an NZW mouse (short scrapie incubation period). Includes most of exon 2 of murine Prn-p gene. |
