The insert has an internal HindIII site about 170 bp from the vector HindIII site. The insert also contains DdeI sites (2), TaqI sites (2), Sau3A1 sites (2) , and a RsaI site. Restriction digests of the clone give the following sizes (kb): PstI--2.7, 0.75; EcoRI--3.4; HindIII--3.3, 0.2. Also available from the ATCC are pBYr5aSP (ATCC 45075) and pBYr5bSP (ATCC 45076) which contain the 0.75 kb PstI fragment cloned in opposite orientations into pSP64. pBYr5 was constructed by random priming. |
