The plasmid contains the following restriction sites (approximate kb from nt 1): BglI--0.24, 1.85; NcoI--0.45; PvuI--0.28, 2.10; PvuII--0.31, 0.67. Expression vector designed for converting restriction sites at the end of inserts. Also permits insert detection by lacZalpha complementation and sequencing from M13 universal and reverse primers. Constructed from pUC18 by modification of the multiple cloning site to include some unique cloning sites surrounded by some direct repeat sites. Pairs of identical restriction sites are in the same reading frame to maintain lacZ' insert detection regardless of the enzyme used. |
