Restriction digests of the clone give the following sizes (kb): EcoRI--19.0, 11.0 (doublet), 4.2; BamHI--23.0, 7.4, 5.8, 4.0, 2.5 1.5; HindIII--23.0, 9.4, 5.8, 4.0, 3.3, 0.96; PstI--14.0, 9.6, 5.3, 3.0, 2.7 (doublet), 2.2, 1.8, 1.2, 0.86, 0.5; AvaI--19.0, 10.0, 6.6, 4.6, 3.1, 2.4, 1.6, 1.2. The allele frequencies of the MspI B1/B2 and the StuI C1/C2 polymorphisms show significant population dependence. The MspI A1/A2 alleles can be detected with a 3.3 kb MspI fragment from the insert. The MspI B1/B2 alleles can be detected with the 5.5 or 5.1 kb MspI fragments from the insert. This clone was isolated from a HaeIII/AluI genomic library in which the DNA was ligated to EcoRI linkers. The sizes of the EcoRI fragments in the insert are (kb): 11.0 and 4.2. Enzyme(s) not detecting polymorphism: AvaII, BamHI, BglII, EcoRI, HincII, HindIII, MboI, PstI, PvuII, RsaI, SstI, TaqI (tested 7 individuals). |
