Restriction digests of the clone give the following sizes (kb): BamHI--3.7; EcoRI--3.7; SacI--3.7. The GAL10 RNA start site is disrupted, thus providing only one direction of transcription. Unless a start site for transcription is provided, little or no message should be made from inserts cloned into the GAL10 side of the promoter cassette. No pBS polylinker sites are included in the GAL1/GAL10 promoter cassette. Plasmid containing a BamHI-GAL1/GAL10-EcoRI cassette of 685 bp useful for making other constructions. The order of the major features in this plasmid is: f1 ori - lacZ' - T7 promoter - Kpn1/MCS/BamHI - GST-GAL1 promoter - GAL10 promoter - EcoRI/MCS/SacI - T3 promoter - lacI - pMB1 ori - ampR. |
